Whole-genome shotgun

One method of preparing a genome for sequencing. The genome to be sequenced is sheared into many small pieces. The ends of this set of small pieces are sequenced (usually by a multiplexed approach). This creates a very large collection of short sequences (~100-1000nt in length). This collection is then assembled into a genome using a variety of approaches:

• de novo
• reference genome
• syntenic path assembly
